چکیده

Photosystem II particles that retain the ability to evolve 0 2 have been used to examine acceptor and inhibitor sites in the photosynthetic electron transfer chain between Q and plastoquinone. Employing the water to dichlorobenzoquinone reaction to assay photosystem II activity, we have demonstrated that electron transport in thylakoids and particles is equally sensitive to inhibition by DCM U, dinoseb, metribuzin, HQNO and DBMIB. Based on differential sensitivity to inhibition by DC M U vs. HQNO or DBMIB, we suggest that when synthetic quinones, e.g. 2,6-dichlorobenzoquinone operate as Hill reagents in particles they are reduced primarily by the plastoquinone pool. When synthetic quinones, e.g. 5,6-methylenedioxy-2,3-dim ethyl benzoquinone act as autoxidizable acceptors they accept electron from the Q /B com plex at a point that is located between the DCM U and HQNO (DBMIB) inhibition sites.

Electron spin resonance in zero magnetic field of triplet states of chloroplasts and subchloroplast particles.

Recently, the new technique of electron spin resonance in zero magnetic field, as observed by Microwave Induced changes in Fluorescence (MIF) has been applied to the study of the light-induced triplet state in chloroplasts under reducing conditions [ 1 ] . Two resonances were detected at 723 and 952 MHz. The resonances were observed also in samples in which the acceptor of photosystem 2 was red...

Superoxide- and Ethane-Formation in Subchloroplast Particles: Catalysis by Pyridinium Derivatives

Oxygen reduction by chloroplast lamellae is catalyzed by low potential redox dyes with E'0 values between -0 .38 V and -0 .6 V. Compounds of E'0 values o f -0 .67 V and lower are inactive. In subchloroplast particles with an active photosystem I but devoid of photosynthetic electron transport between the two photosystems, the active redox compounds enhance chlorophyll bleaching, superoxide form...

The electron donor side of photosystem II: the oxygen evolving complex.

The 02-evolving complex (OEC) of photosynthesis resides near the inner side of the thylakoid membrane. Currently, it is believed to be composed of at least four polypeptides: (1) an intrinsic 27-34 kilodalton (kD) polypeptide that may be associated with manganese (Mn) ; (2) a peripheral 33 k D polypeptide, perhaps also associated with Mn; (3) a peripheral 24 kD polypeptide; and (4) a peripheral...

Chloramphenicol Mediates Superoxide Production in Photosystem II and Enhances Its Photodamage in Isolated Membrane Particles

Chloramphenicol (CAP) is an inhibitor of protein synthesis, which is frequently used to decouple photodamage and protein synthesis dependent repair of Photosystem II during the process of photoinhibition. It has been reported earlier that CAP is able to mediate superoxide production by transferring electrons from the acceptor side of Photosystem I to oxygen. Here we investigated the interaction...

Shortening of life-time of the pair [P680+Pheo-] contributes to general inhibitory effect of dinoseb on electron transfer in PS-II.

It is shown that dinoseb, added to subchloroplast photosystem-II (PS-II) preparations from pea at a concentration higher than 5 microM, along with blocking the electron transfer on the acceptor side of PS-II, induces the following effects revealing its capability to have redox interaction with the components of PS-II reaction center (RC)-pheophytin (Pheo) and chlorophyll P680: (1) acceleration ...
